Description

This human CCL14 ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human CCL14 in multiple samples, including serum, plasma, or tissue homogenates. It also uses the enzyme-substrate chromogenic reaction to visualize and analyze the analyte levels through the color intensity. The intensity of the colored product is in direct proportion to the CCL14 levels in the sample and is measured at 450 nm through a microplate reader.

CCL14, also known as HCC-1, is found in high concentrations in blood plasma and acts as an inflammatory chemokine after N-terminus cleavage. It promotes the chemotaxis of monocytes, eosinophils, and T lymphoblasts. CCL14 specifically binds to CCR1, CCR3, and CCR5 to exert biological effects. CCL14 plays a dual role in the occurrence and development of cancer. A study showed that CCL14 inhibited the proliferation and invasion of colon cancer cells by impeding the formation of M2-like tumor-associated macrophages (TAMs). CCL14 was downregulated in several human cancers, including hepatocellular carcinoma (HCC). CCL14 expression in HCC tissues correlated with prognosis. Low CCL14 expression is linked to poorer overall survival, disease-specific survival, progression-free survival, and relapse-free survival in multiple cohorts of HCC patients, particularly at early disease stages.

It induces intracellular Ca(2+) changes and enzyme release, but no chemotaxis, at concentrations of 100-1,000 nM, and is inactive on T-lymphocytes, neutrophils, and eosinophil leukocytes. Enhances the proliferation of CD34 myeloid progenitor cells. The processed form HCC-1(9-74) is a chemotactic factor that attracts monocytes, eosinophils, and T-cells and is a ligand for CCR1, CCR3 and CCR5.
